Sperm is often indistinguishable or difficult to identify relative to the rest of the sample, especially when being viewed through a brightfield microscope. These manual scans of slides can result in user fatigue and the potential for missing positive samples.
However, the PAXcam Microscope Camera system, used in conjunction with new revolutionary staining techniques for fluorescence microscopy, makes sperm identification and documentation quick, accurate and easy. The dual staining technique uses one stain for cell nuclei in general, and another that is specific for human sperm.
When switching to the sperm-specific stain under fluorescence microscopy, human sperm heads are clearly identified and countable, standing out from the background material due to their unique staining properties.
Brightfield and fluorescent images can be captured from the PAXcam and stored in the PAX-it image database software, where they can be labeled and identified by case number and other user-defined tags, for easy retrieval in the future.
Once the images have been stored, the PAX-it image analysis software can easily count the number of sperm in the field, creating repeatable processes, eliminating human error, and greatly reducing processing and documentation times. Image blending techniques in the PAX-it software allow fluorescent and brightfield images to be overlaid on each other, to show specifically in the field where the human sperm can be found.

Simply select the icon of the saved image from the PAX-it database, select one of the user-created MS Word, Excel or PowerPoint templates of your choice and PAX-it automatically assembles the customized report in seconds. Your forensic reports can include images, annotations, notes, database information, measurement and image analysis data, graphs……anything that you need.

If rape kits are being processed in a batch mode, users may automate the process by using the PAX-it motorized stage control.
